He has no less than six children. → 『彼には6人もの子供がいる』　この no less than ~ は～を強調しています。

He has not less than six children　→　『彼には6人以上の子供がいる』

なおご参考までにless はlittleの比較級で基本的には数えられないものに使いますので、文法的には not (or no) fewer than 6 childrenとすべきで、学校での試験ではご質問の文はダメとされる可能性があります。ただ普通にはless でもアリです。

彼は少なくとも6人子供がいる。
