What do you usually do on rainy days?
の方がいいのでは？

＞人生で一番大切なものは何だと思いますか？
What do you think the most important things in life?

What do you think is the most important thing in life?
isが抜けたのと、最も大切なものが一つだったら、things をthing にし、もし、たくさんある中の一つは何ですか？　と問いたい場合は、

What do you think is one of the most important things in life?
としたらどうでしょう？
