②の場合はオッケーですが、①はいけません。
It is <〇> that I’ve wanted. の形にすべきです。

①では、It is the book that I've wanted. の場合のように、[that I've wanted] が修飾する名詞として [the book] が必要です。

ところが、②の場合の what は、一般的に、what = [ the thing(s) that ]
の役割を持っています。つまり、【「もの」＋ that 】 に置き換えられます。what の中に「もの (thing(s)」が入っていると考えてよいでしょう。
この例だと、It is the thing(s) that I've wanted. の意味です。
