rookie はスポーツチームや軍隊などでは使われますが会社で使う公式用語ではないのでご注意ください。発言者も非常識な人と見られる上に失礼です。

複数の場合：

If the rookies do not fully understand the rule, please teach them the rule once more!

１．いずれにしてもこの会話をしている二人には、新入社員のことも規則のことも既知ですから、"the" をつけましょう。「その」新入社員、「その」規則

２．roukieの綴が間違ってます。

３．one more ではなく、once more (or again)

４． 「あまり＝fully 」を入れるといいでしょう。

５．understand は他動詞なので、about は不要。
