大道路をはさんで南側に位置します

「そのお店は、図書館から大道路をはさんで南側に位置します」
というのを英語で言いたいと思います。

The shop is located in the south side of the library across the main road.

→このinが問題です。こうかかれると、図書館の南側敷地内みたいな感じで、しかも通りの反対にあるとは？？？みたいなことになります。

The store is in the south of the town.
これは町の南の方で、町の中にあるという意味です。
The store is on the south of the town.
これは町の南に接している、つまり、南側の隣りの町との境界線に接している、ちょうど境界線のあたりにあるーーという意味です。
The store is to the south of the town.
これは、町の南の方角で、町の外にあることになります。

以上のようなことですから
The shop is located to the south side of the library across the main road.
とすれば、図書館の敷地を出てさらに南、通りの向こうということで、toが正解となります。
なお、The shop is across the main road, to the south of the library. とやった方がわかりやすいかなと思います。まず、細かい across the main roadを先に言い、その後、おおまかな方向を示すという語順です。
また、大道路が賑やかなお店が並んでいるようなところなら、roadの代りにstreetがよろしいと思います。

　　　　何の問題もなく通じると思いますが、in の代わりに　on　を使うような気がします。
